If you want to access the files on your local machine, then you will want to keep your computer on when you travel. If you only need to access files on the USB drive attached to the Video, then you should only need to keep the Video on.
I just downloaded the Premium Software License - there's a little hiccup because it came with no Activation Code so I ended up installing the free version, notwithstanding the fact that I paid the full $29.95 price via Paypal/Digital River. I submitted a support ticket so I expect this to be resolved momentarily.
My real question is, now that I have both the Desktop Premium Software and the Pogoplug Video on my home network, and I'm travelling overseas for a couple of weeks, should I keep both on at the same time? I know the Pogoplug Video has to stay on, but not sure about the desktop software. But then I might find some exotic data or files overseas and may want to upload them to my home PC. Wouldn't keeping the desktop PC online make it easy to do so 24/7 over several time zones?

P.S. I also noticed that when I installed the Premium Desktop software, my Lacie hard drive attached to the Pogoplug Video vanished from my Windows Explorer file directory. Any idea how to get it back?
If you don't see you drive you may need to uninstall, restart & reinstall.
This is know issue that will be resolved soon.
