Can anyone answer this?

What happens if I can change the name of the destination folder used for an existing Active Copy item?

If I change the name of the destination folder, I assume I will at least need to delete the existing Active Copy task and create a new one -- but will Active Copy need to recopy all the files to the "new" location, or will it read and recognize them as valid copies and not retransfer them all?

You should be able to rename it then reset Active Copy to copy to the new folder.
It will check and see if any of the files in it are newer or different than what is on the originating drive. If not it won't copy them over again.

Thanks Brandon. I finally got around to trying this and you're correct: Pogoplug did not feel the need to recopy all the files.
