In theory, this is possible but probably not recommended. The application would have to be completely self-contained. For a Windows system, this will be difficult since most programs require a local setup procedure in which the registry is modified, files that it needs are installed locally, etc.
It's more likely to work on a Mac or Linux setup, but again, files may be required locally for the applications to work. In addition, the program might run more slowly than it would locally.
All in all, I don't think this is the intended use for Pogoplug but with the Drive software, it's possible in theory. Why don't you give it a shot and let the community know how your experience went?

I am wondering if it possible to install applications on the pogoplug. Not with openpogo. What I mean is can I install an application on my laptop and use the pogoplug as the install location. So as long as I have internet access I can run the application from my laptop without it taking up hard drive space. I want to get a netbook with not much space but install all my applications on my external drive connected through my pogoplug. Is this possible? I could try it I suppose, but thought someone else may have already tried.
mac has the additional issue of requiring certain extended attributes on executables and support files that we don't persist via the drive application.
I personally wouldn't recommend it, cause, even if it does work, it will probably run really slow. Remember, you'd be running the program remotely through your network.
@bcpogo
Have you looked at http://portableapps.com/ ?
They have quite a variety of apps which can run on MS Windows. These apps do not need to be installed and do not (mostly) use the registry. You can carry them around on a usb stick. You could put them on your Pogoplug and they should work reasonably well on your local network but you will be limited by your upload bandwidth if you try to use them remotely.
